INTRODUCTION
Automating complex, multi-step methods requires personnel with laboratory science, robotics programming skills, and design of experiment expertise. They must invest a signi?cant amount of time outlining the work?ow and transferring methods for robotic automation and obtaining such resources can be tasking. Here, we automated thecomplex phosphopeptide enrichment methods starting from crude cell lysate to liquidchromatography-mass spectrometry (LC-MS) ready processed samples as an example.Protein therapeutics continue to expand as a major class of new drug products. Research and development into proteins require a puri?cation processes using a range of resins. Integrated work?ows with IMCStips demonstrate puri?cation of different proteins using different INtip? chemistries and the ?exibility of the Hamilton MicrolabSTAR to achieve high throughput work?ows with minimal human intervention.
?Automated protein A (MabSelect SuRe) INtip puri?cation and buffer exchange in 2 hours.
?Peptide mapping using denaturation, reduction, alkylation and trypsin digestion.
?Examples of modular programs to run for small proof of concept experiments 8-24 samples to larger scale sample processing 96 sample batches.
?Modular pre-templated programs for protein digestion, peptide desalting and phosphopeptide enrichment run in single run or in three separate stages
